Description

Biotin-Ahx-Ub-MTC (UbiQ-378) is an activity-based probe for deubiquitinating enzymes (DUBs). It is based on ubiquitin with an N-terminal biotin and C-terminal methyl hydrazinecarboxylate (MTC) as electrophile. The MTC group has the unique property that it will react with a DUB active site cysteine similarly as a ubiquitinated protein substrate, forming a tetrahedral transition state in which the oxygen anion is stabilized by the anion hole. A 6-aminohexanoic acid (Ahx) linker is used to create extra space between the biotin and Ub protein for efficient access of biotin binding entities.
